Y-chromosome DNA testing is often used when it is not possible to perform a paternity test between the child and the alleged father, and in other complicated family relationships where there is uncertainty about paternity. Y-chromosome DNA testing indirectly establishes whether a child is related to the putative father's brothers or other male individuals who share a common paternal lineage.
In paternal terms, where it is not possible to test the presumed biological father, a Y-chromosome DNA test cannot distinguish the direct family relationship between the tested individuals, but the product does accurately distinguish whether or not the tested individuals share the same paternal lineage.
Example:
If a male test subject and his putative uncle (the putative father's full sibling) are tested, the Y-chromosome DNA profiles must match. If the profiles do not match, the tested putative uncle is excluded.
The Y chromosome is passed down from father to son unchanged through many generations, but although the Y chromosome is only found in males, females can indirectly participate in a Y chromosome DNA test if they are interested in finding out their paternal lineage. Through their biological male family members such as their father, brother, uncle or grandfather, the male line can be clarified.
